How Prolistem® works (the science in brief)
Think of the classical androgen pathway as the main road for sperm production. When it’s blocked, the body can sometimes use a backup road (non‑classical pathways) to maintain sperm production. Prolistem® can trigger the backup road.
The goal is to restart or enhance spermatogenesis, acknowledging that results vary by individual biology and diagnosis.
Prolistem® uses a proprietary compound—glycyrrhizic acid, which converts in the body to glycyrrhetinic acid. This compound inhibits 17β-HSD, an enzyme that drives testosterone synthesis, allowing a temporary hormonal reset.
This reset triggers activation of critical cell-signaling pathways like MAPK, PI3K/Akt, and Notch, which are essential for restarting early-stage sperm development. It’s like pressing a biological reset button—giving the testicles a chance to begin again.
Who is Prolistem® for?
Men diagnosed with non‑obstructive azoospermia, including patterns such as Sertoli Cell Only and maturation arrest.
Men planning micro‑TESE who want to optimize the pre‑surgical window.
Men with certain genetic backgrounds (e.g., mosaic Klinefelter, partial AZFc deletions) may respond; AZFa/AZFb cases typically have lower success. Genetic counseling is recommended.
What to expect & timeline
Duration
Standard course: 6 months.
In some cases of Sertoli Cell Only and/or very high FSH (>30 IU/L), plan for a 12‑month course under clinician guidance.
Monitoring
Monthly semen analysis throughout treatment (see Recommendation Table).
Periodic hormone labs as directed by your clinician.
Do not expect to find sperm before completing the full course.
If no sperm after the full course, Schedule micro‑TESE within 2 months of completion to capture the best window.
- Plan for IVF/ICSI if sperm appears; do not expect natural pregnancy.
